For slides and small welled plates (chamber slides, 24-well, 96-well, etc.)

  • Open packaging carefully to allow the original sleeve to be used for storage
  • Transfer enough collagen to coat the cell culture surface of each well
  • After ~2 minutes tilt the plate slightly and pipette off the excess collagen from each well being careful not to touch the cell culturing surface
  • Transfer the excess collagen back to the stock solution for future use.
  • Allow the plate to dry thoroughly
  • Return to the plates to their original packaging and seal or wrap in saran wrap
  • Store at -20ºC may be stored indefinitely

 

For larger plates (P10, T25, etc.)

  • Collagen will be added to each plate dropwise with a pipette and spread with a sterile bent glass rod.  The amount of collagen required must be determined empirically.  The following amounts have been determined for our P10 plates.
  • Open packaging carefully to allow the original sleeve to be used for storage
  • With a 5ml pipette transfer two or three drops of 1x Rat Tail Collagen to each dish
  • With an ethanol soaked and flame sterilized bent glass rod spread the collagen so as to coat the entire cell culturing surface.  If more collagen is required it may be added.  If an excess of collagen is present it may be removed or allowed to dry on the plate.  In either case note the proper amount of collagen required for future coatings.
  • Allow the plate to dry thoroughly in sterile hood
  • Return to the plates to their original packaging and seal or wrap in saran wrap
  • Store at -20ºC may be stored indefinitely.  Bring dishes to RT just prior to replating cells.
