To differentiate the Cells with NGF:

It is recommended that the cells be fed with a very specific nerve growth factor (see reagents) to ensure good differentiation of the currents for recording.

  • In practice, for the flasks of cells that will be used to record (T25's), I feed the cells with complete media that contains 50 (or up to 200) ng/ml of NGF, 24 hours after plating from a cell passage.
  • After 5-12 days of differentiation in NGF, I replate the cells onto coated plastic/glass coverslips in 35 mm tissue culture plates for recording the next day (day 6-13 for differentiation with NGF).
